Product Introduction

RPMI-1640 is a versatile cell culture medium optimized for suspension cells (e.g., lymphocytes, hybridomas) and select adherent lines (e.g., certain cancer cells). This formulation includes GlutaPlus™ (L-alanyl-L-glutamine) for stable glutamine supply, while excluding HEPES and sodium pyruvate to suit specific experimental needs.

 

Product Specification

Basal Composition:

  • Amino acids, vitamins, inorganic salts.
  • 2.0 g/L glucose (11 mM) – Moderate energy support.
  • Phenol red (pH indicator; optional phenol red-free versions available).

Glutamine Source:

L-alanyl-L-glutamine is a cell culture supplement designed to replace L-glutamine in cell culture media at an equimolar concentration.
This dipeptide exhibits high stability in aqueous solutions and does not undergo spontaneous degradation. Instead, it is gradually cleaved by peptidases secreted by cells, releasing L-glutamine for cellular uptake and utilization.
Unlike L-glutamine (which degrades into toxic ammonia at 37°C), L-alanyl-L-glutamine remains intact in solution, ensuring long-term culture reliability.

Excluded Components:

  • No HEPES – Relies on NaHCO₃/CO₂ buffering (requires 5% CO₂ incubation).
  • No sodium pyruvate – Suitable for metabolic studies or cells with low pyruvate dependence.
  • pH: 7.0–7.4 (adjusted with NaHCO₃).

 

  • Sterility: 0.1 μm filtered.

Supplementation Guidelines

  • Serum: Add 5–10% FBS (or serum-free alternatives like ITS for specialized cultures).
  • Antibiotics: Optional 1% penicillin-streptomycin.
  • Additional Factors: Cytokines (e.g., IL-2 for T cells) or growth factors as needed.

2–8°C, protect from light.
