Agarose is a white or slightly yellow powder, odorless. Hot water is its best solvent, and it can also be dissolved in dimethyl sulfoxide and formamide solutions. Agarose is generally dissolved in water when heated to above 90°C, and forms a good semi-solid gel when the temperature drops to 35-40°C. This is the main feature and basis for its many uses. As the agarose solution cools, the sugar chains of the two agarose molecules wrap around in a double helix to form a three-dimensional network structure. Upon further cooling, the discrete double helices clump together and become tightly packed, forming a stiffer gel. It is often used to prepare nucleic acid analysis gels, such as agarose gels for DNA or RNA electrophoresis.
Procedure of Gel preparation:
1. Prepare an appropriate amount of gel preparation and electrophoresis buffer (commonly used 0.5 x TBE or 1 x TAE buffer);
2. According to the gel production volume and gel concentration, add accurately weighed agarose powder into a glass Erlenmeyer flask containing a certain amount of electrophoresis buffer (the volume of electrophoresis buffer added should not exceed 50% of the capacity of the Erlenmeyer flask).
3. Use plastic wrap or PE gloves to cover the mouth of the conical flask, and then heat to dissolve the agarose in the microwave.
4. When the solution is cooled to about 50°C, a certain proportion of nucleic acid dye (SerRed nucleic acid dye or other nucleic acid dye, etc.) can be added and mixed thoroughly;
5. Pour the agarose solution into the gel making tank, select a suitable comb and insert it into the corresponding position. The gel thickness is generally controlled between 3 ~ 5mm. If subsequent experiments require cutting the gel to recover the nucleic acid in the strip, you can increase the amount of gel appropriately. Thickness, if there are air bubbles in the gel, they need to be driven out.
6. The gel can solidify in about 30 minutes at room temperature (gels with different concentrations have different solidification times, which can be adjusted according to the actual situation). After pulling out the comb, place the gel in the electrophoresis tank for electrophoresis. The electrophoresis buffer should be submerged in the gel. Sample well.
Storage and Shipping Condition:
